ISOLASI OCOTILLON DARI EKSTRAK ETIL ASETAT KULIT BATANG Aglaia Elaeagnoide (A.Juss) Bent dan UJI BIOINSEKTISIDA (ISOLATION OF OCOTILLONE FROM ETIL ACETATE EKSTRACT OF Aglaia Elaeagnoidea (A.Juss.) BENTH’S STEM BARK and BIOINSECTICIDE TEST)

Abstract

Abstrak. Penelitian ini bertujuan untuk menentukan struktur molekul dilakukan dengan metode isolasi dan identifikasi, serta uji bioinsektisida isolat tersebut. Isolasi dimulai dengan ekstraksi serbuk kering kulit batang tumbuhan tersebut dengan pelarut methanol dan dipartisi dengan pelarut etilasetat. Ekstrak etil asetat  difraksinasi dengan metode Kromatografi Cair Vacum (KCV), Kromatografi Kolom Grafitasi (KKG), dan dimonitor dengan Kromatografi Lapis Tipis (KLT). Identifikasi isolat dilakukan melalui spektroskopi UV-Vis, IR, dan GC-MS. Hasilnya dari jenis aglaia ini telah ditemukan senyawa triterpen jenis dammaran yaitu 20,24-Epoxy-25-hydroxy-dammaran-3-one (Ocotillon). Uji bioinsektisida dilakukan dengan variasi ekstrak (EAE), isolat (IAE), dan dibandingkan dengan insektisida sintetik (ISM). Variasi konsentrasi untuk ekstrak 0, 20, 40, 80, 160, dan 320 mg/L, sedangkan untuk IAE dan EAE  0, 2, 8, 16, dan 32 mg/L. Hasil LC50 untuk 24, 48, dan 72 jam berturut-turut untuk EAE 701,273 mg/L;  556,705 mg/L; dan 492,748 mg/L. IAE 64,1378 mg/L; 43,9271 mg/L; dan 33,3234 mg/L. ISM 58,3098 mg/L; 41,8455 mg/L; dan 34,7747 mg/L.

Abstract. The purpose of this research was to determined molecule structure by the method of isolation and identification, as well as isolate of bioinsecticide test. Isolation started by extraction dry powder of plant’s stem bark by using methanol solvent and partitioned with ethylacetate solvent. Then ethylacetate extract was fractionated by Vacuum Liquid Chromatography (VLC), Gravitational Column Chromatography (GCC) which always monitored by Thin Layer Chromatography (TLC). Identification of isolates was determinated by UV-Vis spectroscopy, IR, and GC-MS. Result from Aglaia species have been found dammaran type triterpene compound that is 20, 24-Epoxy-25-hydroxy-dammaran-3-one (Ocotillone). Bioinsecticide test conducted by the variation extract (EAE), isolates (IAE), and compared with synthetic insecticides (ISM). Variation of extract concentration were 0, 20, 40, 80, 160, and 320 mg / L, whereas for IAE and EAE were 0, 2, 8, 16, and 32 mg / L.
