Abstract

Abstrak. Tujuan penelitian ini adalah untuk mengetahui pengaruh asam sitrat dan fitase Bacillus subtilis Holiwood Gresik pada jagung (Zea mays L.) terhadap kadar fitat, daya cerna protein dan bioavailabilitas Zn. Fitase yang digunakan merupakan ekstrak kasar dari isolat Bacillus subtilis HG. Rancangan penelitian yang digunakan adalah Post Test-Only Control Group Design. Tahapan penelitian: 1) untuk analisis kadar fitat jagung dengan variasi bahan perendam berupa asam sitrat 9%, 50 mL/50 g (12 jam) dan fitase 250 µL/50 g  (30 menit). P1 adalah jagung tanpa perendaman (kontrol), P2 perendaman asam sitrat dan P3 kombinasi perendaman asam sitrat, kemudian fitase., 2) untuk analisis daya cerna protein-Protein Efficiency Ratio (PER) pada Mus musculus, digunakan jagung hasil tahap I. Data yang diperoleh dianalisis dengan Anava satu arah (α= 0.05). Hasil: 1) ada pengaruh variasi bahan perendam terhadap kadar fitat jagung. Kadar fitat secara berturut-turut dari P1, P2, dan P3 adalah  11.378, 3.975, and 1.402 (mg/g)., 2) penurunan kadar fitat jagung tahap I dapat meningkatkan PER dan bioavailabilitas Zn Mus musculus. Nilai PER dan % bioavailabilitas Zn secara berturut-turut dari P1, P2 dan P3 adalah 1.9 dan 12.31; 2.5 dan 25.22; 2.7 dan 36.68.

Abstract.  The aim of this research was describe the effect of variation soaking with citric acid and phytase Bacillus subtilis Holiwood Gresik in corn toward concentration of phytate, protein digestibility, and Zn bioavailability. In this research used crude extract of Bacillus subtilis HG isolate. Research design used Post Test-Only Control Group Design. Phases of research: 1) for analyze phytate concentration in corn with variation soaking were citric acid 9%, 50 mL/50 g (12 hour) and phytase 250 µL/50 g (30 minutes). P1 was corn without soaking as control, P2 soaking with citric acid, and P3 combination soaking with citric acid, then phytase., 2) for analyze protein digestibility-Protein Efficiency Ratio (PER) and Zn bioavailability Mus musculus, used corn result phase I. Data were analyze by One-way Anava (α= 0.05). Results: 1) there was effect of variation soaking toward phytate concentration in corn. Phytate concentration from P1, P2, and P3 were 11.378, 3.975, and 1.402 (mg/g) respectively., 2) decreasing concentration of phytate in phase I obtained increasing protein digestibility and Zn bioavailability Mus musculus. PER value and % Zn bioavailability from P1, P2, and P3 were 1.9 and 12.31; 2.5 and 25.22; 2.7 and 36.68 repectively.
