Abstract

Abstrak. Tujuan penelitian ini untuk mengetahui waktu produksi optimum bakteriosin dari Lactobacillus plantarum B1765 berdasarkan aktivitas penghambatannya terhadap Staphylococcus aureus. Uji aktivitas dilakukan menggunakan metode difusi kertas cakram pada setiap fase pertumbuhan, dimana fase pertumbuhan ditentukan dengan mengukur tingkat kekeruhan suspensi bakteri menggunakan spektrofotometer UV-Vis pada λ 560 nm. Hasil penelitian menunjukkan bahwa Lactobacillus plantarum B1765 mengalami tiga fase dalam pertumbuhannya selama 48 jam, yaitu fase lag pada jam 0-6, fase log pada jam 6-18, dan fase stasioner pada jam 18-48. Waktu produksi optimum diidentifikasi sebagai waktu bagi Lactobacillus plantarum B1765 dalam memproduksi senyawa antimikroba bakteriosin secara optimal, yakni ditentukan berdasarkan besarnya zona bening yang terbentuk di sekitar kertas cakram yang berisi supernatan bebas sel Lactobacillus plantarum B1765. Produksi bakteriosin dari Lactobacillus plantarum B1765 dilakukan pada waktu inkubasi 26 jam, dimana terbentuk zona bening dengan diameter terbesar sebesar 13,1375 mm. Sehingga diperoleh aktivitas penghambatan sebesar 2062,259 AU/mL.

Abstract. The aim of this study was to determine the optimum bacteriocin production time of Lactobacillus plantarum B1765 based on its inhibitory activity against Staphylococcus aureus. The activity test was done by disc diffusion method at each growth phase, wherein the growth phase was determined by measuring the turbidity of bacterial suspension using UV-Vis spectrophotometer at λ 560 nm. The results showed that Lactobacillus plantarum B1765 had three phases in its growth for 48 hours, the lag phase at 0-6 hours, the log phase at 6-18 hours, and the stationary phase at 18-48 hours. The optimum production time was identified as the time for Lactobacillus plantarum B1765 to produce an optimal bacteriocin antimicrobial compound, determined based on the clear zone formed around the disk-paper was containing cell-free supernatant of Lactobacillus plantarum B1765. Production of bacteriocin from Lactobacillus plantarum B1765 was done at incubation time 26 hours, which formed the largest clear zone at 13,1375 mm. Thus, the value of inhibition activity is 2062,259 AU/mL.
